The 27th Cavalry Regiment (Colored) was part of the 2nd Cavalry Division (Horse) (Colored), the wartime Black cavalry division activated at Fort Clark, Texas, on February 25, 1943. Assigned to the division's 5th Cavalry Brigade, the regiment trained within a formation that still reflected horse-cavalry organization at a time when the Army's combat needs were shifting toward infantry, armor, and service support.

The division staged at Camp Patrick Henry in February 1944, sailed from Hampton Roads, and reached North Africa on March 9. The 27th Cavalry was inactivated there on March 27, 1944.
